This N-terminal Nrf2 polyclonal antibody detects both polyubiquitinated Nrf2 at 90 kDa and native Nrf2 at 67 kDa. Nrf2 is a transcription factor that forms a heterodimer with a small Maf protein and binds to the antioxidant response element in the upstream promoter region of many antioxidative genes.{22024} Under normal, unstressed conditions, Nrf2 is sequestered in the cytoplasm where it is bound to and ubiquitinated by Kelch like-ECH-associated protein 1 (Keap1) and cullin 3.{22021} Certain inflammatory, environmental, or oxidative stressors disrupt Keap 1 binding and prevent ubiquitination. This leads to a buildup of Nrf2 in the cytoplasm, and subsequent translocation into the nucleus.{22023}
